Hort. Sci. (Prague), 2008, 35(3):95-98 | DOI: 10.17221/6/2008-HORTSCI
The objective of this study was to investigate the possibility of optimizing routine tissue culture methods to proliferate two sweet cherry cultivars Karešova and Rivan. Shoot tips of two genotypes were successfully established in vitro. Six proliferation MS media containing 1, 2 and 4 mg/l BAP (6-benzylaminopurine), 0.5 and 1 mg/l TDZ (thidiazuron) or 10 mg/l 2iP (6-(γ, γ-dimethylallylamino)purine) were tested. The highest proliferation rate (3.0 ± 0.1) was obtained for Rivan on MS medium containing 2 mg/l BAP. In the case of cultivar Karešova, any of the cytokinins tested did not promote satisfactory proliferation. The highest...
Nutrients content and yield in selected cultivars of leaf lettuce (Lactuca sativa L. var. crispa)

Hort. Sci. (Prague), 2008, 35(3):99-106 | DOI: 10.17221/3/2008-HORTSCI
Five cultivars of leaf lettuce (Lactuca sativa L. var. crispa) - Bergamo, Dubáček, Frisby, Lollo Rossa and Redin - were evaluated in two-year experiments carried out at the Faculty of Horticulture in Lednice (Mendel University of Agriculture and Forestry in Brno). Experiments were conducted in two trial years, 1998 and 1999; the lettuce was cultivated in three seasons: spring, summer and autumn. After the harvest, contents of following nutrients were evaluated: vitamin C, minerals (K, Na, Ca, Mg), fibre, dry matter and nitrates. The weight of leaf rosette was also recorded. The contents of selected substances and weights of leaf rosette were ranged...
Bulblet regeneration from ex vitro root explant in lily hybrids

Hort. Sci. (Prague), 2008, 35(3):107-112 | DOI: 10.17221/7/2008-HORTSCI
The influence of growth regulators on in vitro bulblet formation from ex vitro roots was studied in asiatic and oriental hybrids of Lilium The root segments (3-4 mm) isolated from the middle zone of 2-3 cm ex vitro root were cultured on Murashige and Skoog (MS) medium containing 1 or 1.5 mg/dm3 naphthalene acetic acid (NAA) and/or benzyladenine (BA). Bulblets were not produced in the presence of NAA and BA alone. A significant increase in the per cent explants producing bulblets was observed with 1.5 mg/dm3 NAA and 1 mg/dm3 BA. Maximum number of bulblets and average fresh weight per...

Hort. Sci. (Prague), 2008, 35(3):113-129 | DOI: 10.17221/4/2008-HORTSCI
Lettuce (Lactuca sativa) is the most important crop in the group of leafy vegetables. It is characterized by considerable morphological and genetic variation. The crop comprises seven main groups of cultivars (including oilseed lettuce) differing phenotypically; they are usually described as morphotypes. Lettuce breeding is primarilly focused on various morphological features and resistance against diseases and pests. The accurate description of lettuce germplasm provides basic information useful for lettuce breeders. The construction of a lettuce descriptor list has been stimulated by the international genebank community. This list consists...

Hort. Sci. (Prague), 2008, 35(3):130-135 | DOI: 10.17221/661-HORTSCI
Maintenance of garlic (Allium sativum L.) genetic resources has had a long tradition in the Czech Republic. The collection was founded in 1951 and its current holder is the Crop Research Institute, Department of Vegetables and Special Crops in Olomouc. The collection consists of 613 genotypes. The oldest genotype was acquired in 1954. The collection has been evaluated according to the Descriptor list for Allium spp. (IPGRI 2001). 17 characters of the Descriptor list were supplemented with 5 additional characters. The photos of all garlic genotypes were taken. The genotypes were divided into the three main morphological groups according...
